低能N^+辐照提高小麦反转录转座子Wis2-1A的转录并影响其邻近基因表达 被引量:5

摘要 为研究小麦反转录转座子Wis2-1A在低能离子束辐照小麦生物效应中的作用,用实时定量PCR的方法测定不同注量下Wis2-1A的转录水平及在基因组中的拷贝,并用转座子显示技术研究了Wis2-1A在注量为5×1017N+/cm2的低能离子束处理后的转录对其邻近基因表达的影响,结果显示:不同注量的N+辐照都能提高Wis2-1A的转录水平,但在基因组水平上的拷贝没有大的提高,且在较高注量下,由于Wis2-1A的高水平转录导致其邻近的某些基因沉默,这说明Wis2-1A在低能离子注入生物效应中起主要作用。 In order to investigate the roles of Wis2- 1A in the low-energy beam biological effects, the transcription and the copy number in genome of Wis2-1A were determined by real-time quantitative PCR. At the same time, the effect of the Wis2- 1A on the expression of its flanking genes under exposure of the low-energy ion beam was also investigated. The results showed that the low-energy N^+ beam implantation could promote the transcription of the Wis2-1A and restrain the expression of the retrotransposon's flanking genes. It was suggested that Wis2-1A played an important role in responsing the low-energy ion beam biological effects.
