凋亡蛋白Bax和抗凋亡蛋白Bcl-2与胎膜早破的关系

摘要 目的探讨细胞凋亡相关基因蛋白Bax、Bcl-2在胎膜早破(PROM)组与孕足月胎膜未破裂组胎膜组织上的表达情况及与PROM发病因素的相关性。方法PROM孕妇44例(孕36周以后)作为研究组,26例孕足月无胎膜破裂孕妇作为对照组,采用光镜、透射电镜观察2组胎膜上的细胞是否存在凋亡现象。采用免疫组化法检测凋亡相关基因蛋白Bax、Bcl-2在2组胎膜组织上的表达情况。结果bax的阳性率及表达强度在PROM组中高于对照组(P<0.01),Bcl-2的阳性率及表达强度在PROM组中低于对照组(P<0.01)。PROM组44例胎膜组织中Bax、Bcl-2的表达强度无相关性(rs=0.07162,P>0.05)。结论Bax与Bcl-2的表达失衡可能是胎膜组织上的细胞凋亡基因促凋亡的作用机制。 Objective To explore the protein expressions of Bax and Bel-2 in premature rupture of membrane(PROM) and not rupture of mature membrane and their relations with PROM. Methods The membrane tissues of 44 pregnant women with PROM ( research group) and 26 normal full term pregnant women with no rupture of membrane( control group) were collected. First the apoptosis was observed in the above samples using light and transmission electron microscope, then the protein expressions of Bcl-2 and Bax were detected adopting techniques of immunochemistry and histochemistry. Results The positive ratio and expression levels of Bax protein of the pregnant women with PROM was higher than that of control group(P 〈0. 01 ) ,while the positive ratio and expression levels of Bcl-2 protein of the pregnant women with PROM was lower than that of control group ( P 〈 0. 01 ). Among the membrane tissues of 44 cases with PROM, Bax and Bcl-2 expressions did not have a correlation each other( rs = 0. 07162,P 〉 0. 05 ). Conclusions Expression unbalance of both Bax and Bcl-2 may be a mechanism promoting apoptosis by apoptosis genes in the membranes.
