摘要
目的:观察中药红景天对实验性血管性痴呆(VD)大鼠大脑海马中乙酰胆碱酯酶(AchE)活性及神经元细胞凋亡的影响。方法:取健康Wister雄性大鼠,随机分为假手术组、模型组、红景天组、尼莫地平组。除假手术组外,其余各组采用双侧颈总动脉永久性结扎法,造成慢性脑灌注不足所致VD模型。术后红景天组给予红景天5g/kg/d,尼莫地平组予尼莫地平20mg/kg/d灌胃,假手术组与模型组以等量生理盐水灌胃。检测各组大鼠海马组织中AchE活性及海马神经元细胞的凋亡率。结果:海马组织中AchE活性及神经元细胞的凋亡率,红景天组及尼莫地平组与模型组比较,差异有显著性意义(P<0.05);红景天组和尼莫地平组相比,差异无显著性意义(P>0.05)。结论:红景天可能通过降低大鼠海马组织中的AchE的活性和抑制神经元凋亡,对VD大鼠有一定的防治作用。
Objective:To explore the effect of Rhodiola on AchE activity and neuron apoptosis in hippocampus tissue of vascular dementia rats. Methods : 64 male wistar rats were divided randomly as sham operation group, saline control group, Rhodiola group and Nimodipine group. Bilateral common carotid arteries occlusion(BCCAO) was applied to wistar rats for vascular dementia models induced by chronic hypoperfused brain except the sham operation group. Then the rats in Rhodiola group and Nimodipine group were intragastrically given Rhodiola by 5 g·kg· d^-1, and Nimodipine by 20 mg·kg· d^-1, while the rats in simulated control group and saline control group were intragastically given saline of the same dosage. AchE activity and neuron apoptosis in hippocampus tissue were to be determined. Results :The Ache activity and neuron apoptosis in hippocampus tissue of Rhodiola group and Nimodipine group were different significantly from those of saline group(P〈0.05). The difference between Rhodiola group and Nimodipine group was not significant(P〉0.05). Conclusion: Rhodiola might decrease Ache activity and inhibit neuron apoptosis in hippocampus tissue of rats against VD.
