摘要
目的:研究康欣胶囊对兔动脉粥样硬化(AS)血管细胞黏附分子-1mRNA(VCAM-1mRNA)表达的影响。方法:将40只纯种雄性新西兰大耳白兔随机分为模型组、阿托伐他汀钙组(简称他汀钙组)、康欣胶囊低剂量组(简称低剂量组)、康欣胶囊高剂量组(简称高剂量组)、正常对照组,每组各8只。6w后,测定各组血清中总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)及极低密度脂蛋白胆固醇(VLDL-C),进行主动脉血管病理形态学观察,并检测内膜粥样硬化斑块中VCAM-1mRNA的表达水平。结果:与模型组比较,高剂量组血清TC、TG、LDL-C、VLDL-C水平显著下降(P<0.05,P<0.01);模型组血脂明显高于正常对照组。VCAM-1mRNA表达模型组最高,高、低剂量组VCAM-1mRNA表达较模型组下调,有显著性差异(P<0.05),以大剂量组效果为佳。结论:康欣胶囊可明显抑制AS的发生,其机制与减少兔AS血管VCAM-1mRNA表达,抑制AS过程中的炎性反应密切相关。
Objective:To investigate the effects of Kang Xin Capsule on expression of VCAM-1 mRNA in rabbits with atherosclerosis. Methods:40 purebred New Zealand large ear white male rabbits were randomized into model group,atrovastatin (statin)group, Kang Xin Capsule high and low dose groups (high-dose group and low-dose group)and normal control group,8 rabbits in each one. 6w later,serum TC,TG,HDL-C,LDL-C and VLDL-C were measured so as to observe aortic vessel in pathology and morphology and detect VCAM-1mRNA expression in intimia artherosclerotie plaque. Results :Comparing with model group,the levels of serum TC,TG,LDL-C and CLDL-C were reduced significantly in high-dose group (P〈0.05,P〈0.01). The level of blood lipid in model group was higher than that in normal control group. LCAM-1mRNA expression was the highest in model group and down-regulated in high and low dose groups,indicating significant difference(P〈0.05),of which the efficacy in high-dose group was the best. Conclusion:Kang Xin Capsule inhibits atherosclerosis remarkably and the mechanism may be related to reduction of vascular VCAM-1mRNA expression and inhibition of inflammatory response in atherosclerosis.
