摘要
目的研究双羟基黄酮醇(3′,4′-Dihydroxyflavonol,DiOHF)对去势山羊心肌梗死后左心室重构的动态影响。方法雌性山羊经去势处理后,根据临再灌注前静脉用药的不同随机分为DiOHF(2 mg/kg)和赋形剂(VEH)治疗组(n=8/组),实验动物阻断冠状动脉左前降支1 h后施行持续四周的再灌注,左心室内压监测和心脏超声检查分别在实验开始前,再灌注2周和4周进行,实验结束后处死动物,测量心肌梗死面积。结果与基准值相比,再灌注2周和4周后VEH组的左心室舒张末期压,左心室舒张末期容积和左心室收缩末期容积均显著增加(P<0.05);而射血分数和缩短分数则显著减少(P<0.05),DiOHF治疗组不仅逆转了上述变化,而且左心室体积、左室间隔厚度及左室后壁厚度的减少也明显低于VEH组(P<0.05)。此外,与VEH组(80.5±6.9)%相比,DiOHF组(46.2±11.2)%的心肌梗死面积显著缩小(P<0.05)。结论DiOHF治疗对去势山羊心肌梗死后的左心室重构具有明显的阻抑作用。
Objective To study the kinetic influence of 3', 4'-dihydroxyflavonol (DiOHF) on postinfarction left ventricular (LV) remodeling in castrated goats. Methods Following the castration of female goats, animals were randomly divided into DiOHF ( 2 mg/kg) group and vehicle (VEH) group ( n = 8 ) according to the different drugs administered immediately before the reperfusion period. Experimental myocardial infarction (MI) was achieved by occluding the left anterior descending coronary artery for 1 h followed by 4 weeks after reperfusion. LV pressure recording and echocardiogram were performed at baseline, 2 and 4 weeks after reperfusion, and myocardial tissues were collected at the end to determinate infarct size. Results Compared with baseline, LV end-diastolic pressure, LV end-diastolic volume and end-systolic volume in the vehicle group were significantly enhanced following 2 and 4 weeks of reperfusion ( P 〈 0.05 ), which led to considerable decrease ejection fraction and fraction shortening ( P 〈 0.05 ). However, DiOHF treatment reversed those changes, and the reductions in the posterior end-diastolic wall thickness, LV septa thickness and LV mass in DiOHF group were also significantly less than those in vehicle group by the end of reperfusion( P 〈 0.05 ). Additionally, the infarct size in DiOHF treated group (46.2 ± 11.2 )% markedly reduced as compared with that in vehicle treated group ( 80.5 ± 6.9 ) % ( P 〈 0.05 ). Conclusion DiOHF treated daily during the reperfusion period following MI in castrated goats markedly reduces the magnitude of post-infarction LV remodeling.
