摘要 目的:观察水通道蛋白-3(AQP-3)在小鼠肾集合管发育过程中的表达,探讨其与小鼠肾集合管发育的关系及作用。方法:免疫组织化学SABC染色、免疫荧光技术结合体视学方法以及免疫印迹技术检测胎鼠及仔鼠肾AQP-3的表达。结果:AQP-3在胚龄14 d的胎鼠可见微弱的表达,其表达部位为集合管主细胞的基侧面膜。体视学测量和免疫印迹均显示AQP-3从胚龄14 d的胎鼠开始表达,随着胚龄的增加,其表达量逐渐增加,到生后1 d,达到最高水平,此后其表达量几乎维持在较高水平。结论:推测AQP-3在小鼠肾发育中对水平衡具有重要的调节作用,对尿浓缩能力完善的限制性作用不大。 Objective: To observe the expression of aquaporin-3 (AQP-3) in the mice kidney during development and maturation, and to discuss the role of AQP-3. Methods: Immunohistochemistry, immunofluorescence and Western blot combined with stereological methods were used to observe and measure the expression of AQP-3. Results: The expression of AQP-3 was faint at embryonic day 14 and obvious thereafter, h was localized at the basolateral membrane of principal cells of collecting ducts. The results of stereological analysis and Western blot suggested that the expression of AQP3 at the basolateral member was increased gradually from embryonic day 14 to neonatal day I and did not change remarkably thereafter. Conclusion: The results suggested that AQP-3 was very important for water balance during the development of the mice renal collecting duct.
