摘要
采用原生质体融合,EMS与LiCl复合诱变,紫外照射与LiCl复合诱变,紫外、EMS及LiCl复合诱变等方法对季也蒙毕赤酵母(Pichia guillermondii)和休哈塔假丝酵母(Candida shehatea)进行育种,以期得到能代谢木糖提高乙醇生产能力的突变菌株。结果表明:通过以上育种方式得到五个乙醇产量较高的菌株XX2,X1,X4,X12和X13,物理化学联合诱变后得到的X1,X4,X12,X13的乙醇产量分别比对照提高了30.9%,15.7%,84.9%,47.1%,融合子XX2与对照相比,乙醇产量提高了37.7%。高产突变株均来自出发菌株Candida shehatea,Candida shehatea选育后没有得到高产乙醇的突变菌株。
The main objectives of the experiment were to select the fermenting xylose, Pichia guillermondii and Candida shehatea to improve the ethanol yield by protoplast fusion, composite, mutagenesis of EMS and LiCI, composite mutagenesis of UV irradiation and LiCI, composite, mutagenesis of IN, EMS and LiCI. The results showed that five mutants of XX2, X1, X4, X12 and X13, which had higher yield of ethanol by fermenting xylose, were obtained. The ethanol output of XX2, X1, X4, X12 and X13 increased by 37.7% ,30.9%, 15.7%, 84.9%, 47.1% respectively comparing with the control. They are all from Candida shehatea .
