摘要
目的研究经聚偏氟乙烯(PVDF)膜分级分离后地龙匀浆液产物的纤溶活性。方法以新鲜红地龙的匀浆液为分离对象,采用不同孔径的聚偏氟乙烯膜进行超滤,对超滤液进行蛋白浓度、分子量分布和纤溶活性的测定。结果截留分子量(MWCO)为50 000的膜对地龙匀浆液的超滤结果最好,其截留液蛋白浓度大,纤溶活性明显高于其它孔径的超滤产物。结论用聚偏氟乙烯膜分离地龙匀浆液,能得到蛋白浓度高,纤溶活性强的产物,可为临床提供高活性的抗血栓药物。
OJECTIVE To investigate into the fibrolytic activity of pheretima homogenate fractionally separated by polyvinylidene fluoride membrane. METHOD With fresh pheretimaa homogenate as the object of isolation and different apertures of polyvinylidene fluoride membrane, determination was carried out on protein concentration, molecular weight distribution and fibrolytic activity. RESULT Polyvinylidene fluoride membrane with a cutoff molecular weight of 50,000 proved to produce the best ultrafiltration for pheretima homogenate. The cutoff fluid had high concentration of protein, and the fibrolytic activity was markedly higher than products of filtration using other membranes with different apertures. CONCLUSION The use of polyvinylidene fluoride membrane in the isolation of pheretima homogenate produces products with high concentration of protein and high fibrolytic activity, which can provide highly active anti - thrombus medicines.
