ERK在SACC-LM及SACC-83细胞中表达水平与活性的比较

摘要 目的:探讨细胞外信号调节激酶(ERK)在涎腺腺样囊性癌细胞中的活性及表达变化。方法:采用RT-PCR和Western blotting法检测SACC-LM及SACC-83细胞系中ERK蛋白mRNA和蛋白表达,测定ERK酶活力。所得实验数据采用SPSS12.0统计软件进行t检验组间比较。结果:ERK在SACC-LM及SACC-83细胞系中的平均活性分别为54.846 pmol·min-1·μg-1和40.281 pmol·min-1·μg-1,SACC-LM细胞中ERK具有较高活性。SACC-LM细胞中ERK有较高表达。结论:ERK蛋白活性变化及蛋白表达与SACC的转移有关。 Objective: To investigate the extracellular signal-regulated kinase (ERK) expression between two cell lines in adenoid cystic carcinoma of the salivary glands. Methods: ERK expression and enzyme activity in SACC-LM and SACC- 83 cell lines were evaluated through RT-PCR and Western blotting technique. Experimental data were analyzed by ANO- VA followed by t test. Results: Activity of Et/K in SACC-LM cell line (54.846 pmol·min^-1·μg^-1) showed significant difference with SACC-83 cell line (40.281 pmol·min^-1·μg^-1), (P〈0.01); expression of ERK within SACC-LM cells (0.834±0.026) also had significant difference with ERK within SACC-83 cells (0.581±0.072), (P〈0.01). Conclusion: ERK activity and expression variations within the SACC may possibly related to tumor invasion.
