二硫化碳对大鼠睾丸组织的影响及维生素E的干预作用 被引量:6

摘要 目的探讨二硫化碳(CS2)对雄性大鼠生殖功能及睾丸组织中铜、锌、钙、镁水平的影响,同时观察维生素E对其的干预作用。方法36只雄性Wistar大鼠随机分为6组,以不同浓度CS2(0、50、250、1250 mg/m3)静式吸入染毒,共10周,另设CS2(1250 mg/m3)+维生素E(250 mg/kg)组和维生素E组(250 mg/kg)。染毒结束后,检测大鼠生殖毒性指标,测定睾丸铜、锌、钙和镁水平。结果各染毒组睾丸脏器系数、附睾重、睾丸横径、附睾尾精子总数及精子活动率均低于对照组,畸形精子率则高于对照组;睾丸组织铜、锌和镁含量而随染毒浓度增加而降低的趋势。维生素E干预后,精子畸形率降低,睾丸横径、精子总数、精子活动率及睾丸铜、锌和镁均有增加,与高浓度组比较差异有统计学意义(P<0.05)。结论维生素E可干预CS2的睾丸生殖毒性。 Objective To study reproductive toxicity of carbon disulfide (CS2) and the levers of copper, zinc, calcium and magnesium of testicles in the male rats and the intervention of vitamin E(VitE). Methods 36 male rats were randomly divided into six groups. Four groups were exposed to CS2 at different concentrations(0, 50, 250, 1 250 mg/ms , breath). The rest two groups were treated with 1 250 mg/m^3 (CS2) plus VitE(250 mg/kg,diet) and VitE (250 mg/kg,diet) respectively. After lO-week treatment, rats were sacrificed and reproductive toxicity criteria of male rats were measured. The contents of copper, zinc, calcium and magnesium in testes were determined. Results Compared with the control group, the testicle coefficient, epididymal weight, sperm count, sperm motility, the contents of copper, zinc and magnesium of testicles decreased significantly, the sperm malformation rate increased. Pathologic changes of testicles were aggravated with the dose of CS:. After intervened with VitE, the parameters were significantly increased (the amount of the sperm malformation rate decreased) compared with the high dosage group( P 〈 0.05). Conclusion The antioxidant VitE significantly protects against on lipid peroxidation in testes of male rats with CS2.
