摘要
目的观察氯化镉(CdCl2)处理人肝癌SMMC-7721细胞株某些生化指标的变化。方法利用四甲基偶氮唑蓝(MTY)法检测细胞存活率;利用生化法检测CdCl2处理人肝癌SMMC-7721细胞株丙二醛(MDA)含量和超氧化物歧化酶(SOD)及谷胱甘肽过氧化物酶(GSH.px)活力。结果不同浓度的氯化镉作用J2、24和48h,随着给予CdCl2剂量的增加,细胞存活率显著下降。随着给药时间的延长,对细胞抑制作用增强,存在着剂量.效应关系、时间-效应关系;给予细胞10~40gmol/lCdCl2作用24h,随着给镉剂量增加,细胞的MDA含量增加,SOD和GSH—px的活力下降。结论CdCl2可致人肝癌SMMC-7721细胞株存活率下降和某些氧化和抗氧化生化指标改变有关。
Objective To expore the effects of cadmium chloride on certain biochemical indexes of human cancer SMMC-7721 cell line. Methods MTT assay was applied to observe the inhibitory effects and biochemical kits were used to determinate the MDA contents and SOD and GSH-px activities in SMMC-7721 cell line treated with cadmium chloride. Results At the doses of 10, 20, 30 and 40μmol/l cadmium chloride, SMMC-7721 cells were treated for 12, 24 and 48 h, the inhibitory rates of SMMC-7721 ceils increased significantly with the increasing dosage and times of exposure in comparison with control group. There exists an obvious time-effect and dose-effect relationship. At the time of treatment 24 h, the content of MDA were increased and the activities of SOD were decreased and the activities of GSH-px were decreased in comparison with control group. There were a significant differences between the cadmium groups and control group( P 〈 0.05). Conclusion The cell growth inhibition induced by cadmium chloride could be associated with the changes of certain oxidative and antioxidative biochemical indexes.
