摘要
目的观察D-limonene(右旋柠烯)对大鼠结直肠癌的生长和E-cad(E-钙黏蛋白,E-cadherin)、β-cat(β-连环素,β-catenin)表达的影响,从分子学角度探讨其抗肿瘤的机制。方法皮下注射致癌剂二甲基肼(dimethylhydrazine,DMH)构建大鼠原发肠癌模型,中期后给予D-limonene干预,免疫组化染色观察癌组织E-cad、β-cat的表达情况。结果E-cad和β-cat在两组均有异常表达,治疗组异常表达率低;β-cat对照组异位表达增加。结论D-limonene降低了E-cad、β-cat异常表达率,可能抑制了肿瘤的发展和转移。
Objective To observe the impact of D-limonene on the growth of colorectal cancer in rats and on the expressions of E-cadherin (E-cad), β-catenin (β-cat) and to explore its anti-tumor mechanism from the perspective of molecular. Methods Subcutaneous injection of carcinogenic agents of DMH were used to construct the primary model of colorectal cancer, D- limonene interference was performed in the medium-term. The expression of E-cad, β-cat was observed by immunohistoehemical staining. Results Abnormal expression of E-cad and β-cat was detected in both treatment and control groups. The abnormal expression rate was lower in treatment group and the ectopic expression rate of β-cat in the control group was higher. Conclusion D-limonene reduced the E-cad, β-cat abnormal expression rate and may inhibit tumor development and transfer.
