摘要
目的观察海风藤提取物对大鼠脑缺血再灌注(I/R)后大脑皮质缺血灶周围区caspase-3蛋白、凋亡诱导因子时相表达及神经细胞凋亡的影响。方法应用线栓法建立Wistar大鼠大脑中动脉阻塞(MCAO)再灌注模型,TTC染色观察梗死灶的分布,应用原位末端标记(TUNEL)和免疫组化技术分别观察模型组、海风藤提取物组和二甲基亚砜(DMSO)组大脑中动脉栓塞90min再灌注6、24、72h时缺血灶周围脑区caspase-3蛋白、凋亡诱导因子(AIF)和TUNEL阳性细胞的时相表达。结果①脑I/R后在缺血灶周围区均出现三者的表达,且表达有几乎一致的规律性。②缺血90min再灌注24、72h,海风藤组与其他两组比较,caspase-3、凋亡诱导因子阳性细胞和TUNEL染色阳性细胞的数量均明显减少(P<0.01)。结论①AIF、caspase-3参与了局灶性脑缺血后神经细胞的凋亡过程。②海风藤提取物可以减少AIF、caspase-3蛋白的表达,减轻迟发性神经元死亡,对缺血性脑组织有保护作用。
Objective To investigate the effect of piper futokadsure extract on the expression of apoptosis-inducing factor (AIF), caspase-3 protein and neuronal apoptosis in perifocal areas after ischemia-reperfusion (I/R) in rats. Methods The I/R rat model was estab- lished by middle cerebral artery occlusion (MCAO) and reperfusion. Infarction zones were confirmed by 2,3,5-triphenyltetrazoliumchloride (TIC) staining. The expression of AIF, caspase-3 positive cells and variations of neuronal apoptosis in perifocal regions injured by ischemia for 90 min, then reperfusion at 6, 24, 72 h points in model, piper futokadsure extract and DMSO groups were detected by immunohistochemistry and in situ apoptotic detection (TUNEL method). Results (1) The expression of AIF, caspase-3 and Tunel positive neurons had the similar laws in perifocal areas at different time points after I/R. (2) The numbers of AIF, caspase-3 and Tunel positive neurons in piper futokadsure group were significantly decreased compared with those in model and DMSO groups at 24, 72 h points in perifocal areas after I/R(P 〈 0. 01 ). Conclusions (1) AIF and caspase-3 may participate in the neuronal apoptotic process after focal cerebral I/R. (2) Piper futokadsure extract may play protective function for the cerebral tissues after I/R by reducing the expression of AIF, caspase-3 and neuronal apoptosis.
