生脉解毒通络胶囊对实验性糖尿病大鼠心肌超微结构的保护作用 被引量:4

摘要 目的研究中药生脉解毒通络(SJT)胶囊对链脲佐菌素(STZ)糖尿病(DM)大鼠心肌局部血管紧张素Ⅱ(AngⅡ)含量和超微结构的影响及相关机制。方法STZ诱导建立DM模型,成模2w后分别灌服依那普利和SJT胶囊低、中、高剂量连续12w,于第14周末应用放射免疫法检测心肌局部AngⅡ含量,并在电镜下观察心肌超微结构的变化。结果与正常对照组比较,DM模型组心肌局部AngⅡ含量明显增加(P<0.01),SJT胶囊中、高剂量治疗组及依那普利对照组均可使DM大鼠心肌局部AngⅡ含量下降(P<0.05、P<0.01),并可使电镜下心肌超微结构得到改善,低剂量组无显著效果。结论SJT胶囊可对DM大鼠心肌超微结构损害起到保护作用,其作用机制可能与抑制DM大鼠心肌局部AngⅡ的激活有关。 Objective To investigate the effects of Shengmai Jiedu Tongluo (SJT) capsule on myocardial local angiotensin (Ang)Ⅱ and ultrastructure in diabetic rats induced by streptozocin (STZ). Methods Diabetes rat model was established by intraperitoneally administering of STZ, and medicated through gastrogavage with enalapril and SJT capsule of large, median and small dose respectively, for successive 12 w, started from 2 weeks after modeling. Levels of myocardial Ang Ⅱ were detected by immunoradiometry and changes of myocardial uhrastructure were observed by an electronic microscope. Results AngⅡ level in model group was significantly higher than that in control group ( P 〈 0. 01 ). In enalapril and SJT capsule of large and median dose groups, levels of Ang Ⅱ was lower significantly compared with that in the model rats (P 〈0. 01, P 〈0. 05). The myocardial uhrastructure changes were also improved in SJT capsule of large and median dose groups, but not in small dose SJT capsule group. Conclusions SJT capsule may partly suppress myocardial local levels of Ang Ⅱ and decrease significantly the myocardial injury of diabetic rats by reducing the level of myocardial Ang Ⅱ.
