喘可治对急性肺损伤大鼠预防作用的研究 被引量:3

摘要 目的探讨喘可治注射液对急性肺损伤(ALI)的预防作用。方法采用腹腔注射脂多糖(LPS)复制ALI大鼠,随机将25只大鼠分为喘可治小剂量组、中剂量组、大剂量组、模型组和正常组,每组各5只。观察每组支气管肺泡灌洗液(BALF)中总蛋白含量、白细胞计数及分类、动脉血氧分压(PaO2)和肺组织湿/干重比(W/D);进行外周血正常组T细胞亚群检测;采用ELISA法检测肺组织干扰素γ(IFN-γ)、白细胞介素8(IL-8)、巨噬细胞炎性蛋白2(MIP-2)含量;通过肺组织切片、HE染色进行病理形态学观察。结果模型组大鼠出现呼吸窘迫和低氧血症,肺水肿严重,W/D比值显著升高(P<0.05),肺组织IFN-γ、IL-8、MIP-2浓度、BALF中蛋白含量以及中性粒细胞百分比亦显著升高(P<0.01)。与模型组相比,喘可治不同剂量组呼吸窘迫和低氧血症症状均有不同程度改善,肺水肿减轻,W/D比值和肺组织IFN-γ、IL-8、MIP-2浓度以及BALF中蛋白含量、中性粒细胞百分比较模型组均显著下降(P<0.01或P<0.05),CD3、CD4T细胞、CD4/CD8显著升高(P<0.001)。结论喘可治注射液防治ALI的作用可能是其能减少IFN-γ、IL-8、MIP-2等细胞因子的释放以及调节细胞免疫功能共同作用的结果。 Objective To study the preventive effects of CKZ injection in rats with acute lung injury (ALI). Methods ALI models were induced by peritoneal injection of lipopolysaccharides (LPS). Twenty-five SD rats were randomly divided into five groups (n = 5) : small dose CKZ group, middle dose CKZ group, large dose CKZ group, model group and normal group. Total protein level, white blood cell count and differentiation in BALF, PaO2 and lung wet/dry weight (W/D) were detected or measured. Numbers of T cell subsets in peripheral blood were determined. The concentrations of IFN-γ, IL-8, macrophage inflammatory protein-2 (MIP-2) in lung tissues were detected by ELISA method. Pathomorphology was observed by tissue section and HE staining. Results Respiratory distress and hypoxemia were found in model group, with severe pulmonary edema and significantly higher W/D ( P 〈 0.05). Besides, the levels of IFN-γ, IL-8 and MIP-2 in lung tissues, the concentrations of protein and neutrophils in BALF were significantly higher than those of normal group ( P 〈 0. 01 ). In CKZ groups, respiratory distress and hypoxemia were relieved to some degree, with moderate pulmonary edema. Meanwhile, W/D, levels of IFN-γ, IL-8 and MIP-2 in lung tissues, and concentrations of protein and neutrophils in BALF were significantly lower than those in model group (P 〈 0.01 or P 〈 0.05). The levels of CD3, CD4 T cells and CD4/CD8 were significantly higher than those in model group(P 〈 0. 001 ). Conclusion The preventive and therapeutic effects of CKZ injection in ALI were the results of both reducing the release of cytokines such as IFN-γ, IL-8 and MIP-2 and regulating the cellular immune function.
