P53负调控分子PACT的研究进展

摘要 睾丸P53相关细胞蛋白(P53 associated cellular protein-testes derived,PACT),又称为相关增殖潜能蛋白(proliferation potential protein related,P2P-R)和成视网膜瘤结合蛋白6(retinoblastoma binding protein 6,RBBP6),是为数不多的能够同时与生物体内最重要的两种抑癌基因p53和Rb结合的蛋白之一,参与细胞分化、前体mRNA剪接、周期阻滞、凋亡等一系列重要的细胞事件。自其不同形式的同源体被发现至今,不同实验室对其在不同角度进行了研究,一些功能已经初见端倪。本实验室近期制备了PACT基因敲除的小鼠模型,揭示了PACT在胚胎发育中的不可或缺性,进一步明确了PACT对P53调控机制。本文通过总结不同实验室对PACT的研究工作,确定了PACT的主要结构区域,综述了当前其主要研究方向和进展。 P53 Associated cellular protein-testes derived (PACT), also named proliferation potential protein related (P2P-R) or retinoblastoma binding protein 6 ( RBBP6), belongs to a small subfamily which could interact with both 1)53 and Rb in vitro and in vivo. It could participate in diverse cellular processes, such as apoptosis, cell cycle control, pre-mRNA splicing. During the last ten years, progress has been made in the functional investigation of PACT. Most importantly, the PACT gene knock-out mice model was established and the essential role of PACT in embryogenesis was discovered recently in this laboratory. Furthermore, the molecular mechanism by which PACT controls P53 activity has been elucidated.
