轴突生长抑制因子DNA疫苗对大鼠脑缺血后神经再生的影响

Effect of DNA Vaccine against Neurite Growth Inhibitors on Neural Regeneration Following Focal Cerebral Ischemia in Rats

摘要 目的研究大鼠轴突生长抑制因子的DNA疫苗(pcDNA3.1(+)-neurite growth inhibitors,pcDNA-NGIs)对局部脑缺血神经再生的影响。方法采用永久性阻断大脑中动脉血流的方法制备雄性SD大鼠左侧局部脑缺血模型,分别在阻断血流前后经腓肠肌注射pcDNA-NGIs免疫动物,每周一次,共6周;血清中检出特异性抗体6周后,立体定向脑内注射生物素化葡聚糖胺追踪皮质红核投射的新生轴索。结果无论是阻断血流前还是阻断血流后给予pcDNA-NGIs,局部脑缺血后皮质红核投射的代偿性新生轴索都明显增多。结论本研究提示,阻断血流前或阻断血流后给予pcDNA-NGIs均可提高局部脑缺血后的神经再生。 Objective To elucidate the effect of DNA vaccine against neurite growth inhibitors, pcDNA3.1(+) neurite inhibitors (pcDNA-NGIs) on neurite regeneration following focal cerebral ischemia. Methods The left focal cerebral ischemia was permanently induced by the occlusion of the left middle cerebral arteries in 72 rats. pcDNA-NGIs was injected into the tibial muscle once a week for 6 weeks before or after the stroke. The newly generated axons of corticorubral projection were traced by stereotaxic guided injection of biotinylated dextran amine 6 weeks after the presentation of the specific antibodies in the serum. Results pcDNA-NGIs administrated before or after the stroke led to increase in compensatory new growth of axons of corticorubral projection following the focal cerebral ischemia. Conclusion It is suggested that pcDNA-NGIs administrated before or after the stroke is beneficial to neurite regeneration following the permanent local cerebral ischemia.
