摘要
目的研究FTY720对人心房肌细胞电生理特性及功能的影响。方法取20例心脏手术患者右心耳,将1 Hz的场刺激作用于心房肌组织,采用传统微电极技术记录FTY720对基础状态下心房肌和0.2μmol/L异丙肾上腺素(ISO)处理后心房肌收缩力和动作电位。结果FTY720对基础状态的心房肌动作电位和收缩力无显著影响;但可使ISO处理后的心房肌动作电位时程缩短、幅度降低、收缩力减弱(P均<0.05)。结论FTY720可明显抑制ISO条件下的心房肌动作电位和收缩力,为探讨其作为心脏疾病治疗药物的可能性及其作为免疫抑制剂应用可能带来心血管的副作用提供了一定理论依据。
Objective To investigate the effects of FTY720 on action potential (AP) and contraction force (CF) of human atria fibers. Methods The human atria fibers of right appendages were obtained from 20 patients with cardiac dis ease and paced at 1 Hz,then were treated by FTY720 and isoprenaline( ISO), AP and CF of which were recorded using standard intracellular microeleetrode technique. Results FTY720( 1.4 μmol/L) had no significant effects on the AP and CF of human atria fibers, but significartly inhibited the enhancement effects of 0.2 μmol/L ISO on the CF and AP amplitude (APA) ( P 〈 0.05 ), meanwhile, FTY720 ( 1.4 μmol/L) further shortened the action potential duration (APD) after the atria fiber treated by 0.2 μmol/L I:30 ( △ APD50 30 ms) . Conclusions FTY720 may obviously reverse the -adrenoceptor-induced enhancement of CF and APA.
