摘要
为寻找一种廉价、快捷的方法选育转谷氨酰胺酶(MTG)菌株,采用平板稀释法分离土壤放线菌,根据转谷氨酰胺酶催化反应结果的特性,设计了一种蛋白质交联—絮凝沉淀性能测定方法对分离菌株进行复筛,用比色法测定MTG酶活,并用插片法进行复筛菌株的初步鉴定。结果表明:从土壤中分离出16株放线菌,经复筛后有6株放线菌产生沉淀,即具有MTG酶活性,且其中LH-011的MTG酶活最大,达0.76u·mL-1。经初步鉴定,6株放线菌均属于放线菌科链霉属。可见,用蛋白交联—絮凝沉淀性能测定的方法可以筛选出产MTG菌株。
In order to find a low-cost and ,;imple method for filtrating transglutaminase strains(MTG), plat dilution method, a protein cross-linked- flocculation precipitate method that had been designed according to the characteristic of the transglutaminase catalyzing reaction, slide cultivation and co3orimetry were used to select actinomycetes initially. Afterwards, identify primarily and assay MTG activity respectively. 6 strains had acquired were identified primarily as Streptomyces spp..The highest MTG activity was 0.76u.mL^-1. It was suggested that a protein cross-linked-floeculation precipitate method could be adopted to breed the MTG strain.
