摘要
目的:通过检测尿脱落细胞的hTERT mRNA和端粒酶的活性,探讨建立膀胱癌早期诊断和术后检查的无创方法。方法:利用可提高灵敏度的巢式PCR技术,对尿脱落细胞中hTERT的mRNA进行RT-PCR检测,以TRAP方法加以验证。结果:28例膀胱癌患者尿脱落细胞hTERT的mRNA表达阳性率为93%(26/28)。20例泌尿系良性疾病组患者则为10%(2/20),TRAP方法有很好的一致性。结论:尿脱落细胞hTERT mRNA的检测敏感性高,特异性好,有望应用于膀胱癌的早期诊断和术后复发的无创检查。
Objective: To study the mRNA of human telomerase reverse transcriptase (hTERT) in exfoliated urinary cells of bladder cancer and to evaluate its clinical significance. Methods: We examined the expression of mRNA of hTERT in exfoliated urorhelial cells of bladder and control by RT-PCR. Meantime, the telomere reoeat amplification protocol (TRAP) was performed for indentify the result of mRNA of hTERT. Results: The positive rate of hTERT mRNA of bladder cancer group is 93% (26/28) and control group is 10% (2/20). The result was consistent with the TRAP. Conclusion: The result suggest the expression of hTERT mRNA can be useful in screeing for bladder cancer and diagnosing bladder cancer recrudesce after operation.
