摘要 目的:研究银耳多糖对肝癌HepG-2细胞增殖的影响。方法:采用高温浸提的方法提取银耳多糖,并进行提取定量;将银耳多糖作用于肝癌HepG-2细胞,采用台盼蓝排斥试验测定细胞活力和生长曲线。结果:在分别培养1 d、2d和3 d后,银耳多糖干预各组的活细胞率都低于对照组(P<0.001)。生长曲线法显示,银耳多糖干预各组的细胞倍增时间延长,银耳多糖对增殖细胞的杀伤率最高达89.29%(>80%),且存在剂量依赖关系。结论:银耳多糖对肝癌HepG-2细胞具有直接抑制作用。 Objective:To investigate the effects of polysacharides from tremella on the growth of human hepatoma cell line HepG-2. Methods : The polysaccharides was extracted from tremella fucifornis berk by high temperature. After determining the content of the products, the HepG-2 was cultured with the extract, and trypan blue dye exclusion test waa conducted to examine the cell growth by growth curve. Results: After being cultured at 1 d, 2 d and 3 d, respectively, the cell viable count in groups with tremella polysaccharides was lower than that of the control group(P〈0.001). The growth curve showed that the cell multiplying time in groups with tremella polysaccharides delayed with an anti-tumor rate of 89.29% and dose-dependent effect. Conclusion: Tremella polysaccharides can kill the cell HepG-2 of human hepatoma.
