摘要
目的研究iASPP在乳腺癌细胞株和乳腺浸润性导管癌中的表达情况,为乳腺癌的诊断治疗和预后提供新的指标。方法设计引物采用RT-PCR技术分别扩增5种人乳腺癌细胞株(MDA-MB-231,MCF-7,Bcap-37,HBL-100和ZR-75-30)和乳腺浸润性导管癌组织中iASPP的mRNA,应用Quality One软件分析iASPP扩增产物的相对含量。免疫组化染色判断iASPP在乳腺癌细胞株和乳腺浸润性导管癌中的表达情况。结果iASPP基因只在乳腺癌细胞株Bcap-37、MCF-7和HBL-100中表达,在其它2株细胞中不表达;iASPP mRNA在58例乳腺浸润性导管癌组织中有51例表达iASPP(87.9%),癌旁组织不表达iASPP。免疫组化染色显示iASPP蛋白在58例乳腺浸润性导管癌中有53例表达(91.4%),而在对应的癌旁组织中不表达iASPP。结论iASPP mRNA在乳腺癌细胞株中的表达具有差异性;iASPP蛋白在乳腺浸润性导管癌中表达,有助于浸润性导管癌的诊断,有望成为乳腺浸润性导管癌患者治疗的新靶点。
Objective iASPP is a kind of apoptosis-stimulating protein Of p53 (ASPP) family members include. This study was to detect the expression of iASPP in 5 kinds of breast cancer cell lines and breast infiltration duetal cancer tissue, so as to provide the marker for diagnosis and prognosis of breast cancer. Methods The cross mRNA of 5 kinds of breast cancer cell lines and breast cancer tissue was extracted with tripure reagent and the iASPP gene was amplified by RT-PCR, and then the average optional density of RT-PCR products was assayed by Quality one soft-ware. The expression of iASPP protein was detected with immunohistochemistry. Results The mRNA of iASPP was expressed in Beap- 37, MCF-7 and HBL-100 cell lines, but not in MDA-MB-231, ZR75-30 cell lines. The iASPP protein was expressed in breast cancer tissue (53/58) and not in pancancer tissue. Conclusion The expression of iASPP is different from each other in 5 breast cancer cell lines. The iASPP protein can be used as a marker for diagnosing the breast cancer and will be used as a new target for treatment of breast cancer.
