摘要
对从新疆盐碱地土壤样品中分离出的一株嗜碱菌株XJ21进行鉴定,提取其胞外多糖,研究多糖的活性。用平板分区划线法分离菌株,通过形态学、生理、生化和16SrDNA测序对菌株进行鉴定。从发酵液中提取胞外多糖,通过Sephadex G-100凝胶柱层析对多糖进行分离纯化。小鼠迟发型变态反应(DTH)测定免疫活性,Ames试验测定抗突变活性。结果:嗜碱菌株XJ21为克劳氏芽胞杆菌,命名为Bacillus clausiiXJ21。其胞外多糖对环磷酰胺所致免疫低下小鼠迟发型变态反应有恢复作用;对NaN3所致的鼠伤寒沙门菌TA100的突变有抑制作用。
To isolate and identify an alkalophilic strsain from salina soil in Xinjiang and to extract its exopolysaccharide, the strain was isolated by streak-plate, and identified by biophysical and biochemical tests and sequence of 16Sr DNA. The polysaccharide was extracted from broth and separated by sephadex G 100. DTH test was used to investigate immune enhancing activity and Ames test was used to study the anti-mutagenic effect. Experiment result showed that the alkalophilic strain XJ21 was isolated and identified as Bacillus clausii, named as Bacillus clausii XJ21. Its exopolysaccharide could promote the DTH response of immuno deficiency mice caused by cyclophospamide and inhibit the mutation to Salmonella typhimurium TA100 induced by NAN3.
