摘要
目的:探讨复方虎杖胶囊(Fufang hu zhangjiao nang,FHZ)对高尿酸血症小鼠T淋巴细胞亚群的影响。方法:小鼠随机分为空白组、别嘌呤醇组、模型组及5.0%、2.5%、1.25%FHZ组,灌胃给药6d后,采用腹腔注射黄嘌呤一次造模,半小时后摘眼球取血,尿酸酶比色法测定血尿酸;处死小鼠,取脾脏,剪碎、研磨后,制备成单细胞悬液,单色法应用流式细胞仪测定CD3+、CD4+、CD8+百分比及CD4+/CD8+比值。结果:模型组血尿酸明显升高(与空白组比较,P<0.05),而别嘌呤醇组和5.0%、2.5%、1.25%FHZ组能明显降低血尿酸(与模型组比较,P<0.05)。应用流式细胞仪检测脾脏T淋巴细胞分型结果显示:模型组、别嘌呤醇组和空白组CD3+、CD4+、CD8+及CD4+/CD8+差异均无统计学意义;且5%FHZ组与模型组、空白组、别嘌呤醇组相比较差异亦无统计学意义。结论:FHZ具有明显的降低高尿酸血症小鼠血尿酸的作用,优于别嘌呤醇。急性高尿酸血症和FHZ对脾脏T淋巴细胞亚群无明显影响。
Objective:To study the influence on the subpopulations of T lymphocyte in mice administrated Fu fang hu zhang jiao nang(FHZ).Methods:Adopt 10% xanthine i.p.in mice to duplicate hyperuricemia model,and administrate drugs for 6 days beforehand.Then,to detect serum urate concentration by uricase chromatometry and the percent of CD+3、CD+4、CD+8 and CD+4/CD+8 ratio of spleen T lymphocytes by monochromic flow cytometry(FCM).Results:5.0%,2.5%,1.25% FHZ group and allopurinol group had significant differentiation respectively compared with model group(P<0.01,n =8),and were lower than model group,respectively.And 5.0%,2.5% FHZ group had significant differentiation respectively compared with allopurinol group.It denoted that different dosage FHZ can respectively reduce serum urine content in hyperuricemia of mice,and even be lower than normal level.In contrast,there were no significant differences between allopurinol group,model group and normal group in the percentage of CD+3、CD+4、CD+8 cells and CD+4/CD+8 ratio in spleen lymphocytes,nor were there significant differences between normal group,model group,allopurinol group and 5% FHZ group in the percent of CD+3、CD+4、CD+8 cells or CD+4/CD+8 ratio of spleen lymphocytes.Conclusion:FHZ can obviously reduce serum urine concentration.But acute hyperuricemia in mice and FHZ do not have significant influence in spleen T lymphocyte subpopulations
