摘要
目的:探索一种提取大鼠冻存脂肪组织总RNA的有效方法。方法:在Trizol分离试剂说明书基础上,参考文献提取脂肪组织RNA的方法,离心(10000rpm,10min)去除上层脂类液体后才开始抽提。结果:用此法提取的大鼠冻存脂肪组织总RNA,经电泳鉴定及进一步RT-PCR检测,仍未能显示出较高纯度和完整性。结论:提取大鼠冻存脂肪组织总RNA的方法还需进一步改进。
Objective： Finding an effective extraction method of total RNA from cryopreservation rats fat tissue. Method： According to the specification of Trizol reagent, referring to the extraction methods of total RNA from fat tissue in references, Before chloroform extraction , removing the oil of the upper layer by centrifuging（10000rpm, 10min）. Results ： After electrophoresis identification and analyzed by RT-PCR of RNA, the total RNA by this extraction method still can not satisfy experimental requirement. Conclusion ： The extraction method of total RNA from cryopreservation rats fat tissue need to further improvement.
