摘要
探索了一种分离纯化嗅鞘细胞(Olfactory Ensheathing Cells,OECs)的简单易行的新方法,将OECs与合成材料聚羟基乙酸-L-赖氨酸-乳酸(简称PLGK)以及聚乳酸共培养,比较并筛选合适的神经修复材料。通过水接触角测试、材料对细胞黏附率测试、台盼蓝染色,计算材料表面细胞存活率以及扫描电镜观察材料表面细胞生长状况;结果得到了纯度达90%,数量达到3×106的嗅鞘细胞,共培养后测试结果表明PLGK与聚乳酸(PDLLA)比较具有更好的亲水性、细胞黏附能力和存活率以及良好的细胞生长状况。
In this paper, a simple method of purifying OECs was developed. After cultivating the purified OECs onto the surface of synthesized materials of PDLLA and PLGK, the contact angles of materials were determined, the adherent rate and activity rate was calculated, and the growth features on the material surface was examined by scan electron microscope. The resuits shows that an inexpensive method of purifying OECs has been developed and approximately 3 ×10^6 cells which purity is ≥90 % are yielded. PLGK could show better hydrophile, affinity and activity of cells and well cell growth condition contrasting to PDLLA.
