摘要
目的建立高效液相色谱法测定青蒿中山柰素的含量,并对青蒿不同部位的山柰素含量进行比较。方法岛津VP—ODS色谱柱(150mm×4.6mm,5μm),乙腈-0.2%磷酸溶液(35:65)为流动相,测定波长367nm,柱温30%,流速0.8ml/min,进样量20μl。结果山柰素在0.0218—0.436μg范围内呈现良好的线性关系(r=0.9999),平均回收率为98.70%,RSD为1.20%。结论该方法简便可行,重复性好,专属性强,可用于青蒿中山柰素的含量测定。测定结果表明,青蒿叶中山柰素的含量最高,花次之,根较少,茎最低。
Objective To establish HPLC method fbr the determination of kaempferol in Qinghao and analyze kaempferol content in different parts of Qinghao. Methods Kaempferol in Qinghao was determined by HPLC with VP- ODS column (150 mm× 4.6 mm, 5μm). The mobile phase was Acetonitrile -0.2% H3PO4 (35: 65) solution. The detection wavelength was 367nm. The column temperatare was 30℃ , the flow rate was 0.8 ml/min, and the sampling volume was 20μl. Results Linearity of Kaempferolwas good (r = 0.9999) in range of 0. 0218 - 0. 436μg. The average recovery was 98. 70%,RSD was 1. 20%. Conclusion The method is simple, accurate, repeatable and suitable for the determination of kaempferol in Qinghao. The results show that the highest content of Kaempferol is in leaf,flowers the second,less roots,stems the minimum.
