中药复方红苓肝宝对酒精致小鼠骨丢失的预防作用

摘要 目的观察中药复方红苓肝宝对酒精致小鼠骨丢失的预防作用。方法昆明种小鼠50只随机分组,除对照组外,各组予50%(V/V)酒精按4 g.kg-1.d-1灌胃,药物预防组分别在酒精灌胃前给予中药低剂量(HLGB-L),高剂量(HLGB-H)和西药对照,60 d后处死取左侧胫骨上段不脱钙骨切片进行骨组织形态计量学参数测量,腰椎及股骨测定骨生物力学指标。结果酒精组小鼠骨小梁面积百分数(%Tb.Ar)明显降低(-66%,P<0.05),骨形成率(BFR/TV)降低(-35%,P<0.05);腰椎抗压性能和股骨颈结构强度均明显下降。与酒精组比较,HLGB-H组%Tb.Ar增加(+68%,P<0.05),BFR/TV增加(+42%,P<0.05),腰椎抗压性能和股骨颈结构强度增加(分别为+50%,+13%,P<0.05),与低剂量组比有统计学差异,但没达到正常组的水平。结论HLGB-H对酒精摄入造成的小鼠骨丢失有一定的预防作用,不但增加其骨量,改善骨的微观结构,还增强骨的力学性能,但不能完全恢复至正常小鼠的水平。 Objective To study the protective effects of Hongling Ganbao (HLGB) on bone loss induced by alcohol in mice. Methods Fifty six - week - old KM mice, were randomly divided into 5 groups except for the control group. 50% (V/V) alcohol was given to the model mice according to the dosage of 4g/( kg · d). Low dose (HLGB - L)and high dose of HLGB (HLGB - H) were given to mice respectively before the alcohol was consumed in preventive groups. After 60 days, all mice were killed and their left tibial metaphysis were processed in undecalcified sections for bone histomorphometric analysis. The tested biomeehanical properties of vertebra and femccr were tested. Results Compared with normal group, cancellous bone area (% Tb. Ar) was decreased obviously( -66% ) in alcohol group, bone formation rate BFR/TV was decreased( -35% , P 〈 0.05) (and bone resorption parameter Oc. N/Tb. Pm tended to increase) , bone biomechanics properties of femur were decreased significantly. Compared with alcohol group, indexes of % Tb. Ar and BFR/TV of HLGB - H group were increased( + 68% , + 42% respectively) , and bone biomechanics properties including compressivw resistibility of vetebra and structure strength of femur neck both improved significantly. There had statistical significance between TCMC and HLGB - L, but it didnt reach the normal level. Conclusion HL- GB - H could prevent bone loss induced by alcohol by improving bone microstructure and strengthening bone biomechanics properties.
