摘要 建立了PAN载药纤维中有机溶剂N,N-二甲基乙酰胺(DMAc)残留量的反相高效液相色谱检测方法:色谱柱为Kromasil 100A C18,流动相为甲醇/水(体积比28/72)混合溶液,流速为0.8 mL/min,进样量为5μL,检测波长为220 nm,柱温为室温。结果表明:DMAc浓度在5~200μg/mL,色谱峰面积与质量浓度之间线性关系良好,相关系数0.999 2,回收率为98.5%~100.6%,相对标准偏差为0.87%~1.12%。该方法简单、准确,测定所制备的载药纤维含DMAc 579μg/g。 a method of determining the organic solvent of N, N-dimethylacetamide (DMAc) residue in drug-loaded fiber was established by reversed-phase high-performance liquid chromatography. The measurement conditions were decided as followed: Kromasil 100A C18 chromatographic column, methanol/water mixed solution ( volume ratio 28/72) as mobile phase, flow rate 0. 8 mL/min, injection volume 5 μL, wavelength 220 nm, room temperature. The results showed that the chromatographic peak atea had a good linear relationship with the mass concentration, the correlation coefficient was 0.999 2, the recovery rate 98.5% - 100.6% and the relative standard derivation 0.87% - 1.12% when the DMAc concentration was 5 - 200μg/mL. The method was simple and accurate. The DMAc content in the obtained drug-loaded fiber was 579μg/g.
