摘要
目的:为开展骨组织工程研究,建立成骨细胞与支架材料的体外复合培养模型。方法:取新生24h内的Wistar大鼠颅盖骨培养成骨细胞,并将成骨细胞以5×103/孔接种到HA/CS,HA/CS-SF两种支架材料表面常规培养,用倒置显微镜观察细胞形态,通过MTT比色法测出两种支架材料对细胞增殖的影响。结果:培养的成骨细胞呈短梭形或多角形,碱性磷酸酶染色镜下可见胞浆中棕色至黑色颗粒的为阳性细胞。通过MTT chromatometry比色法测定表明HA/CS-SF对细胞增殖有更好的作用。结论:HA/CS-SF具有作为骨组织工程支架材料的可行性。
Objective:To carry out bone tissue engineering research, and to establish a model to provide theoretical bases for the application of scaffold materials in the repair of bone loss. Method:The crania osteoblasts were obtained from newborn Wistar rats within 24 hours. The osteoblasts were transplanted to HA/CS and HA/CS-SF at a density of 5 × 10^3/mL. The biological characteristics of the osteoblasts were observed under the phase-contrast microscope, and the cell proliferation was detected by the assay of MTT. Results :The cell shape was triangle or polygon, aad black particles could be observed in the endochylema of the cultured ceils with ALP stain. The MTT chromatometry showed that HA/CS--SF had a better effect on the cell proliferation. Conclusion:HA/CS-SF is feasibie as the scaffold materials of bone tissue engineering.
