rhIFN-a、rhIL-2与rhTNF-a体外协同抑制SMMC-7721细胞的敏感性实验 被引量:1

摘要 目的:本研究旨在探讨干扰素-a(IFN-a)、白细胞介素-2(IL-2)、肿瘤坏死因子(TNF-a)单一、协同以及与化疗药物DDP的联合作用杀伤肝癌SMMC-7721细胞株的影响。方法:应用四甲基偶氮唑盐比色法(MTT比色法)检测与化疗药物DDP单独或联合应用组及药物作用的12h与24h的光密度值(OD),计算杀伤率。结果:单独应用三种细胞因子时,IFN-a、TNF-a对肝癌SMMC-7721细胞株有细胞毒作用,IL-2对肝癌SMMC-7721细胞株的细胞毒作用不明显。IFN-a、IL-2、TNF-a可以增强DDP的抑瘤作用,与DDP单独作用具有显著性差异(P<0.01)。IFN-a、IL-2、TNF-a联合作用具有协同作用,均与单一应用具有显著性差异(P<0.01)。结论:细胞因子均可以增强顺铂(DDP)的抑瘤作用并与其作用的先后及时间具有依赖关系。IFN-a、TNF-a、IL-2与DDP联合具有协同作用,为肝癌病人化疗提供了理想的参考方案。 Objective:To investigate the effects of rhIFN-a, rhIL-2 and rhTNF-a on the SMMC-7721 cell line in vitro in a single or cooperative form or in combination with chemotherapeutics DDP. Methods: Chemosensitivity of the cells to doxorubicin was analysed by using the MTT assay at 12h and 24h after use of rhIFN-a, rhIL-2 and rhTNF-a alone and in combination with chemotherapeutics DDP. Results :When the three factors were used alone,IFN-a or TNF-a had toxic action on the liver cancer SMMC-7721 cell line, and when they were used cooperatively, there was significant difference in the effects on the cell line (P 〈 0.01). All the cytokines had the enhancement of DDP anti-tumor activity in vitro with the dependence of the anti- tumor effect on their action order and time. Conelusion:rhIFN-a,rhIL-2 and rhTNF-a have the synergistic inhibition in combination with DDP,which provides a better chemotherapy choice for patients with liver cancer.
