摘要
利用组织培养技术,研究了不同pH和NAA浓度对沙枣愈伤组织的影响,结果表明:0.5mg/L与1mg/L NAA浓度对沙枣愈伤组织存活率影响显著,pH对沙枣愈伤组织的存活率有极显著影响.通过方差分析可知,沙枣在pH为5.5时与pH为7.0,8.5,10.0时的存活率有极为显著的差异,在pH为8.5,10.0的盐碱胁迫下也能形成愈伤组织.通过继代培育,筛选出耐盐碱的沙枣优良品种,可为盐碱地生态经济树种的选择和开发利用提供理论依据.
This paper researched the effect of different pHs and concentration of NAA on the formation of callus of Elaeagnus angustifolia L. by using the technology of tissue culture. The results show that the effect of NAA with 0.5mg/L and 1mg/L on livability rate of callus of Elaeagnus angustifolia L is prominent,and the effect of pH on livability rate of callus of Elaeagnus angustifolia L. is apparently higher. There was significant difference of the livability rate of Elaeagnus angustifolia L. between pH 5.5 and pH 7.0,8.5, 10.0. The saline-alkali stress can still form callus when pH is 8.5, 10.0. Therefore,we can cultivate and filtrate the breed of Elaeagnus angustifolia L. ,which can provide some academic basis for the choice and development of the ecological species for the saline.
