ARDS状态下大鼠肺泡Ⅱ型细胞水通道蛋白1表达的变化 被引量:5

摘要 目的观察大鼠ARDS状态下Ⅱ型细胞水通道蛋白1(AQP1)表达的变化,为ARDS发病机制的研究提供新的实验和理论依据。方法运用免疫组化、Western bloting和图像分析技术,对正常大鼠和ARDS大鼠Ⅱ型细胞AQP1的表达水平进行对比观察,分析变化。结果Western blot分析显示ARDS大鼠肺泡Ⅱ型细胞AQP1蛋白条带较对照组明显减弱,免疫组化及Western blot经图像分析结果为ARDS状态下Ⅱ型细胞上水通道蛋白1的表达较对照组有明显的下降(P<0.05)。结论AQP1对呼吸系统水转运可能起到重要作用。 Objective To investigate the alteration of AQP1 in alveolar type Ⅱ ceils of ARDS rats in order to provide new experimental and theoretic evidence in the study of the pathogenesis of ARDS. Methods The expression of AQP1 was assessed with immunocytochemistry, western bloting and image morphometry in oleic acid group and control group. Results Western bloting and image morphometry revealed a significant decrease in AQP1 protein levels in alveolar type Ⅱ cells of ARDS rats compared with control rats. Conclusion The expression of AQP1 in the type Ⅱ cells of ARDS rats significantly decreases as compared with control group( P 〈 0.05 ). Decreased expression of AQP1 in injury lungs suggests that AQP1 may play a important role in fluid transportation.
