摘要
目的:探讨VEGF在人脑正常组织、胶质瘤及瘤周水肿组织中的表达及意义。方法:应用免疫组化SP法检测VEGF在66例人脑胶质瘤组织中的表达。测算EI,进行统计分析。结果:VEGF典型阳性表现为血管内皮细胞的细胞浆及星形胶质细胞、反应性星形胶质细胞和肿瘤细胞的细胞浆和/或突起呈黄色或棕黄色,较少表达于神经元细胞,神经胶质中表达不明确。VEGF在不同病理级别胶质瘤间的表达不同,其表达程度与病理级别相关;而在胶质瘤瘤周水肿组织中的表达与来源肿瘤的病理级别、性别、年龄均不相关。EI与肿瘤病理级别、高低级别、患者年龄有一定的正相关,与VEGF表达、性别、肿瘤发生侧别不相关。幕上胶质瘤的EI与肿瘤病理级别、高低级别有很强的正相关,而与VEGF表达、性别、年龄、肿瘤发生侧别不相关。结论:VEGF在人正常脑组织中异质表达;在人脑胶质瘤肿瘤及瘤周水肿组织中表达增强,并与胶质瘤病理级别相关。瘤周水肿程度与肿瘤恶性度密切相关。
Objective:To study the VEGF expression in the human normal brain tissue, glioma tumor and peritumoral edema tissue. Methods: Immunoh istochemical stains (SP) were performed to examine the expression of VEGF in 66 samples collected (DAB coloration). Expressing color shade was graded into no staining, yellow, bully and brown; expressing scope was distinguished into no-staining, scattered, limited and widespread. So, combining color shade and scope, expressing strength was graded into four classes presented with " -, +, + +, + + + ". Calculated edema index (EI) by measuring preoperational imaging. Applying SPSS 11.5 statistical package, carry out two - and several - samples nonparameter test, and rank correlation analysis on results. P value less than 0.05 was considered statistically significant. Results: VEGF expressed typically in endothelial cytoplasm, cytoplasm and enations of tumor cells, active astrocytes, some gliocytes, but less in neuron and unidentifily in glioma. VEGF expressional degree in glioma, being different between different pathogrades, correlated with pathological grade, but VEGF expressional degree in edema tissue around glioma was not related to glioma pathograde, gender and age so on. EI has a positive correlation with pathograde, high or low grade and age, but not with VEGF expressional degree, gender and tumorigenesis side. Supertentorial glioma EI has a significant positive correlation with pathograde and high or low grade but not with VEGF expressional degree, gender, age and tumorigenesis side. Conclusions: VEGF expressions, the difference between different normal brain tissues, in human glioma and peritumoral edema tissue were stronger than normal, and had correlation with pathological grade. The peritumoral edema degree of human glioma had close correlation with tumor grade.
