康脑液对脑缺血再灌注损伤后神经细胞干细胞因子mRNA表达的影响

摘要 目的在药物干预下观察内源性神经细胞干细胞因子(SCF)mRNA在大鼠脑缺血再灌注损伤后不同时间、不同部位的表达情况。方法成年SD大鼠,以线栓法建立大脑中动脉缺血再灌注模型,随机分为模型组、假手术组和药物干预组。原位杂交技术检测脑缺血1.5h再灌注1~14d后,脑组织不同部位的SCFmR-NA表达情况。结果脑缺血再灌注后,药物干预组、模型组SCFmRNA的表达在皮质区及纹状体区均明显高于假手术组,于7d达高峰,14d下降。结论脑缺血再灌注后SCF mRNA表达在脑内不同部位、不同时间具备同样的规律,可能具有促进内源性神经干细胞的增殖作用。但两组SCF mRNA的表达差异无统计学意义。 Objective To observe the influence of Kang Naoye on the expression of stem cell factor (SCF) mRNA after focal cerebral ischemia-reperfusion injury of different times and different positions in rats. Methods A middle cerebral artery occlusion (MCAO) model was established with the intraluminal suture method. Adult SD rats were randomly divided into model group, medicine intervention group and sham-operation group. The expression of SCF mRNA after 1.5-hour ischemia and 1-day-14-day reperfusion was measured by in site hybridization in different positions of brain. Results After focal cerebral ischemia-reperfusion, the level of SCF mRNA expression in medicine intervention group and model group was obviously higher than that in sham-operation group in the cortical area and corpora striatum area, and peaked at 7 days and dropped at 14 days. Conclusion The expression of SCF mRNA had the same rule in different positions of rats' brain after focal cerebral ischemia - reperfusion. This medicine may promote the effect of proliferation of endogenous neural stem cells. Two groups of SCF mRNA expression was of no significant difference.
