摘要 目的研究丙烯酰胺(ACR)对新生大鼠肾上皮细胞增殖分化的影响。方法利用新生大鼠肾上皮细胞进行原代培养,观察不同剂量ACR(0.1,1.0,10.0,50.0,100.0μg/ml)对细胞形态学及细胞生长与分化过程影响,同时测定蛋白质含量、丙二醛(MDA)含量和超氧化物歧化酶(SOD)活性。结果ACR处理各组细胞体积小,组集落存活率分别为对照组的97%,86%,80%,56%,38%;蛋白质相对含量分别为对照组的92%,90%,85%,53%,33%。ACR剂量为10.0,50.0,100.0μg/ml时和对照组比较,差异均有统计学意义(P<0.05,P<0.01),并显示剂量-效应关系;ACR处理各组MDA含量有升高的趋势,SOD活性有下降的趋热,ACR剂量为10.0,50.0,100.0μg/ml时与对照组比较,差异均有统计学意义(P<0.01,P<0.05)。结论ACR能抑制新生大鼠肾上皮细胞增殖和分化,可能与其能抑制蛋白质合成、DNA有关。 Objective To study the effect of acrylamide on proliferation and differentiation of rat kidney epithelial cells. Methods The primary culture of kidney epithelial cells of new born rat were separated, and digested after purification. Acrylamide was added at the dose of 0.1, 1.0, 10.0, 50.0,100.0 μg/ml, respectively. The cell culture was observed with cytomorghology and cell counting. Concentration and the content of protein, the MTT, malondialdehyde (MDA) and activity of superoxide dismutase (SOD) were determined. Results Acrylamide inhibited differerentiation of the cells by affecting cell proliferation. The inhibition rate was 97 %, 86 %, 80 %, 56 % and 38 %, respectively. The content of protein was 92 %, 90 %, 85%,53% and 33% of that control group, respectively. The results showed that acrylamide (10.0, 50.0, 100.0 μg/ml) could inhibit differentiation of kidney epithelial ceils without affecting cells proliferation. The level of MDA in groups treated with acrylamide showed increasing tendency. The activity of SOD was lower than that of control group, with a significant decreasing in the 10.0 μg/ml group, 50.0 μg/ml group and 100.0μg/ml group. Conclusion The inhibition of acrylamide on rat kidney epithelial cells might be associated with the impact of protein and DNA synthetis.
