摘要
目的将不同数量牙周韧带细胞(PDLC)植入慢性实验性牙周缺损内,观察细胞植入数量与牙周组织再生量的关系,确定可促进牙周组织再生的有效细胞浓度和有效细胞数量。方法人工构建4只Beagle犬慢性实验性牙周缺损模型(5 mm×5 mm×3 mm的牙槽骨U型缺损),分别将载有1×104、1×105和1×106PDLC的胶原膜植入牙周缺损内。12周后行组织学观察,测量新生牙骨质(NC)长度百分率和新生牙槽骨(NB)面积百分率并行统计学分析。结果各组均可见不同程度牙周组织再生,1×104PDLC组和1×105PDLC组与对照组比较差别无统计学意义,1×106PDLC组的NB面积百分率较对照组显著增高。结论在实验性牙周缺损内植入PDLC,可促进牙周组织再生,其有效浓度为5×107mL-1,有效数量为1×106PDLC。
Objective The aim of this study was to compare the regeneration effect of seeding different amount of autologous periodontal ligament cells(PDLCS) into canine artificial furcation defects. Methods Twenty-four experimental class II furcation defects in four Beagle dogs were created surgically. Guide tissue regemeration(GTR) combination with the expanded PDLCs which were seeded in collagen membrane at initial concentration of 5 ×10^5 cells/mL, 5 ×10^6 cells/mL, or 5 ×10^7 cells/mL, and auto- transplanted 1 ×10^4 , 1 ×10^5 , 1 ×10^6PDLCs into experimental class Ⅱ furcation defects, a Bio-Gide membrane alone was implanted into the defects as control. The animals were sacrificed at 12 weeks postsur- gery and periodontal regeneration was determinated by histological and morphometric analysis. Results In both the experimental groups and the control group, there were different extent of regeneration. Histometrie analysis showed that the the 1 ×10^6 PDLCs group were significantly effective than other group. Conclusion Auto-transplantation of periodontal ligament cells is a novel option for periodontal tissue regeneration, and the effective concentration is 5 ×10^7 cells/mL or 1 ×10^6 PDLCs.
