吡格列酮诱导前列腺癌PC-3细胞凋亡的实验研究 被引量:1

摘要 目的研究吡格列酮对前列腺癌(Pca)PC-3细胞凋亡的诱导作用及其可能机制。方法体外培养Pca PC-3细胞,以不同浓度(0,0.1,1,10,100μmol/L)的吡格列酮干预细胞,四氮唑蓝比色法(MTT)和原位细胞凋亡检测法(Tunnel)检验吡格列酮对Pca PC-3细胞的增殖抑制及诱导凋亡作用,流式细胞术检测其对Caspase-3表达的影响。结果10,100μmol/L吡格列酮对前列腺癌PC-3细胞有显著的抑制增殖及诱导凋亡的作用,其干预组Caspase-3表达显著增高,0.1,1.0μmol/L吡格列酮对前列腺癌PC-3细胞无明显作用,其干预组Caspase-3表达表明显变化。结论吡格列酮可诱导Pca PC-3细胞凋亡,作用呈剂量依赖性,其机制可能与细胞内Caspase-3的活化有关。 Objective To research the inductive effects of pioglitazone on apoptosis of PC-3 and its possible mechanism. Methods the cultured PC-3 cells are intervened by pioglitazone of different concen- trations(0, 0.1, 1, 10, 100 μmol/L). MTT, Tunnel methods are adopted to test the inhibitory effects of pioglitazone on PC-3 cell proliferation and its inductive effects on apoptosis of PC-3 cells, meanwhile flow eytometry is employed to test its effects on expression of Caspase-3. Results The pioglitazone of 10 μmol/L and 100 μmol/L showed significant effects on growth inhibition and apoptosis induction on PC-3 cells, and significantly increased the expression level of cellular easpase-3, whereas the pioglitazone of 0.1 μmol/L and 1.0 μmol/L showed no obivious effexts. Conclusion Pioglitazone can induce apoptosis of PC-3 cells with dosage-dependence manner, and its mechanism may be correlated with intracellular activa- tion of caspase-3.
