摘要
目的比较3种传代细胞系(Vero细胞、BHK-21细胞、MDCK细胞)对狂犬病病毒CTN-1V株的敏感性。方法采用混种的方法,将狂犬病病毒CTN-1V株分别接种于上述3种不同来源的连续传代细胞系,在不同时间观察3种细胞的病变情况,同时应用快速免疫荧光灶抑制试验(RFFIT)和ELISA法检测病毒滴度和抗原含量,分析不同来源细胞对狂犬病病毒CTN-1V株的敏感性。结果Vero细胞培养的狂犬病病毒滴度在第8天达到高峰,为7.80LogFFD50/ml,BHK-21细胞病毒滴度在第6天达到高峰,为7.30LogFFD50/ml,而MDCK细胞在第6天病毒最高滴度只有5.55LogFFD50/ml。BHK-21细胞培养的狂犬病病毒抗原含量(A492)值,在各代次均高于Vero细胞和MDCK细胞。结论Vero细胞和BHK-21细胞对狂犬病病毒CTN-1V株的敏感性高于MDCK细胞,且BHK-21细胞产毒高峰比Vero细胞提前。
Objective To compare the sensitivities of 3 continuous cell lines, i. e. Vero, BHK-21 and MDCK cell lines, to rabies virus CTN-1V strain. Methods Inoculate rabies virus CTN-1V strain to Vero, BHK-21 and MDCK cells respectively, observe the CPE on various day after inoculation. Determine the virus titer by rapid fluorescent focus inhibition test (RFFIT) and the antigen content by ELISA. Analyze the sensitivities of cells from various origins to CTN-1V strain. Results The virus titer of CTN-1V strain cultured in Vero cells reached a peak value of 7. 80 LogFFD50 / ml on day 8 and that in BHK-21 cells reached a peak value of 7. 30 LogFFD50 / ml on day 6 after inoculation, However, the peak value of virus titer in MDCK cells was only 5. 55 LogFFD50 / ml. The antigen contents(A492) of rabies virus in BHK-21 cells of various passages were higher than those in Vero and MDCK cells. Conclusion Both the sensitivities of Vero and BHK-21 cell lines to rabies virus CTN-1V strain were higher than that of MDCK cell line. Compared with that in Vero cells, the virus titer of CTN-1V strain in BHK-21 cells reached the peak value early.
