摘要
目的:观察助孕胶囊对PCO模型大鼠血清性激素水平及INH-ACT-FS系统的影响,探讨其对PCOS的治疗作用并分析其可能的作用机制。方法:采用胰岛素联合HCG造模法制备PCO模型大鼠。随机分为对照组、模型组、助孕胶囊组、二甲双胍组。运用放射免疫法测定血清性激素、胰岛素(INS)水平;双抗体夹心酶联免疫吸附法测定血清抑制素(INHB)、激活素(ACTA)及卵泡抑素(FS)的水平。结果:助孕胶囊可明显下调PCO大鼠血清睾酮(T)、促黄体生成素(LH)、INS水平;并能上调血清ACT水平,降低血清INH、FS水平;且INS与ACTA呈负相关,与INHB呈正相关。结论:INH-ACT-FS系统在多囊卵巢的发生、发展中起重要作用;高胰岛素血症与INH-ACT-FS系统有关;助孕胶囊能够通过调整H-P-O轴及影响卵巢局部细胞因子INH、ACT、FS的表达,从而调节生殖内分泌,促进卵泡的发育、成熟及排卵。
Objective: To observe the effects of ZhuYun JiaoNang(ZYJN) on procreate endocrine hormone and inhibinactivin-follistatin system in rat of polycystic ovary (PCO) , analyzing the possible therapeutic mechanism and therapeutical effect. Methods: The PCO rat models were set by insulin combining with human chorionic gonadotrophin (HCG), randomLy divided into four group, which included control group, model group,Chinese crude drug group(ZYJN) and western medicine group(DMBG). The levels of serum gonadal hormone and insulin(INS) were detected by radioimmunoassay. Using the sandwich ELISA technique detected the expressed level of serum InhibinB(INHB), ActivinA(ACTA) and Follistatin(FS). Results: ZYJN could decrease the level of testosterone (T) , luteotropic hormone( LH ), INS, INH and FS in rat serum, but increase the level of ACT in rat serum. It is negative correlation between INS and ACTA, positive correlation between INS and INHB. Conclusion: The inhibin-activinfollistatin system play an important roles in the development of PCO. Hyperinsulinemia is relation with INH-ACT-FS system. ZYJN could regulate hypothalamic-pituitary-ovarian axis and the expressing of ovaries local cytokines such as INH, ACT and FS, then adjusting genesis endocrine and promoting the mature and ovulation of follicular.
