摘要
目的:观察雌二醇(estradiol,E2)对类风湿关节炎(RA)滑膜细胞中核因子κB(NF-κB)p65亚基发生核转位的情况。方法:经关节穿刺术无菌抽取RA患者膝关节液,原代培养获得成纤维型滑膜细胞,用10-6M的E2分别作用1、2、4、8、24h,采用荧光免疫细胞化学法检测滑膜细胞NF-κB p65亚基核转位。结果:荧光显微镜观察染色的滑膜细胞,发现E2处理4、8、24h的滑膜细胞p65发生了核转位,其转位率分别为68%、95%和52%,在8h时达到峰值。结论:雌二醇能诱导NF-κB的活化,这可能是E2参与RA炎症反应的一个重要环节。
Objective: To investigate the nuclear translocation of nuclear factor κB in fibroblast-like synoviocytes controlled with estradiol in rheumatoid arthritis. Methods: Aspirated the synovial fluid from the knee joint of RA patients during therapeutic arthrocentesis. Get the primary culture of fibroblast like synoviocytes from the synovial fluid. Nuclear trans location of the subunit p65 and p50, was observed by indirect immunofluorescence assay when fibroblast-like synoviocytes were respectively stimulated by 10^6μmol/L E2 for 1 , 2, 4, 8 and 24 hours. Results: E2 induced the nuclear translocation of p65 at 4, 8 and 24 hours. The ratio of p65 translocation was 68%,95% and 52%, respectively. Conclusion: E2 could induce the activation of NF-κB,which may play an important role in inflammatory response.
