摘要
目的:研究针刺对多囊卵巢综合征(PCOS)大鼠卵巢转化生长因子α(TGF-α)、表皮生长因子受体(EGFR)表达的影响,探讨针刺促排卵的作用机制。方法:24日龄雌性大鼠颈背部皮下注射脱氢表雄酮(DHEA)的油溶液制作(PCOS)模型,对照组同期皮下注射油剂。PCOS大鼠随机分为模型组和针刺组。模型组不作处理,针刺组大鼠从80日龄起针刺关元、中极、双侧三阴交、双侧子宫穴,1次/天,15min/次,连续6周。治疗结束后各组大鼠断头处死,迅速取血并分离血清,-20℃冰箱保存,待测性激素水平。摘取双侧卵巢,称重,4%多聚甲醛固定,作HE染色和免疫组织化学染色。结果:与模型组相比,针刺组卵巢湿重、卵巢TGF-α、EGFR表达及血清睾酮(T)、雌二醇(E2)水平均显著降低(P<0.01),而卵泡刺激素(FSH)、黄体生成素(LH)、孕酮(P4)水平差异无统计学意义(P>0.05)。结论:针刺能显著降低PCOS大鼠卵巢TGF-α及EGFR的表达,抑制TGF-α对卵巢和激素合成的作用,改善PCOS大鼠多囊样变和高雄激素血症,促进排卵。
Objective:To observe effect of acupucture on expressions of transforming growth factor-α(TGF-α)and epidermal growth factor receptor(EGFR)in ovaries of rats with polycystic ovarian syndrome(PCOS),to explore mechanism of acupuncture effects on ovulation induction.Method:PCOS rats model was experimentally induced by subcutaneous injection of oil solution of dehydroepiandrosterone(DHEA)in immature(24-day-old)female rats for continuous 20 days.Control animals were given with same dose of oil for an equivalent length of time.PCOS rats were randomly divided into model group and acupucture group.The acupoints of Zhongji(CV3),Guanyuan(CV4),Sanyinjiao(SP6)and Zigong(CX-CA1)are selected to perform acupuncture for continuous 6 weeks(15 minutes,once daily)since 80 days old on acupucture group.All the rats were usually killed by decapitation after treatment finished.Trunk blood was collected expeditiously and serum was abstracted for measurement of hormone levels.The ovaries were dissected,trimmed,weighed,and fixed in 4% paraform.Some were stained with hematoxylin and eosin,others were treated with immunohistochemistry stainning method to detect the expressions of TGF-α and EGFR in ovaries of each rat.Results:Compared with model group,the acupuncture group had significantly lower ovary weight(P<0.01),expressions of TGF-α and EGFR in ovaries(P<0.01),as well as serum testerone and E2(P<0.01),but no statistical differences of serum FSH,LH and P4(P>0.05).Conclusion:Acupuncture treatment can conspicuously down-regulate the expressions of serum testerone and E2,as well as expressions of TGF-α and EGFR in ovaries of PCOS rats,inhibit the effect of TGF-α on ovary and hormone synthesis,ameliorate the polycystic morphology of ovaries and hyperandrogenemia,and stimulate ovulation.
