摘要
建立了片剂中美西律含量的高效毛细管电泳安培检测方法。探讨了缓冲溶液种类、浓度、酸碱度及操作电压、进样时间等对检测的影响。以Na2B4O7-NaOH(Na2B4O7浓度为30 mmol/L)为电泳介质,在15 kV高压,pH=8.5的弱碱性条件下,采用柱端安培法检测了美西律的含量。线性范围为5×10^-5~5×10^-2g/L,Y=856.2+123.3c,r=0.999 4,检出限为0.01 mg/L。
Mexiletine hydrochloride was determined by capillary electrophoresis with amperometric detection (CE-AD). The pH, type, concentration of buffer, applied voltage and sample injection time were selected as the investigation variables. Under the optimized conditions:a running buffer composed of 30 mmol/L Na2 B4 07 at a pH value of 8.5, separation voltage 15 kV, injection time 10 seconds and detection potential at 0. 6 V, a linearity exists between peak height(Y) and mass concentration of Mexi/etine hydroehloride(c) in a range of 0. 05 ~50 mg/L, r =0. 999 4, and the determination limit is 0.01 mg/L.
