摘要
目的检测抗神经元核抗体(NeuN)、突触素(Syn)、少突胶质细胞转录因子-2(Oligo-2)及胶质纤维酸性蛋白(GFAP)在中枢神经细胞瘤的表达状况,探讨在中枢神经细胞瘤诊断与鉴别诊断中的作用。方法对中枢神经细胞瘤12例、少突胶质细胞瘤和室管膜细胞瘤各20例,应用EnVision法进行NeuN、Syn、Oligo-2和GFAP检测。结果NeuN和Oligo-2阳性定位于细胞核,Syn和GFAP阳性定位于胞质或胞膜。中枢神经细胞瘤:9例NeuN(+),10例Syn(+),1例Oligo-2(+),4例GFAP(+)。少突胶质细胞瘤:2例NeuN(+),1例Syn(+),13例Oligo-2(+),3例GFAP(+)。室管膜瘤:3例NeuN(+),2例Syn(+),3例Oligo-2(+),15例GFAP(+)。结论中枢神经细胞瘤中NeuN和Syn呈高表达,Oligo-2和GFAP呈低表达,对中枢神经细胞瘤的诊断和鉴别诊断具有应用价值。
Objective To investigate neuronal nuclear antigen (NeuN), synaptophysin, oligodendroglial cell transcription factor- 2 (Oligo-2) and GFAP protein expression in independent sets of central neurocytomas, oligodendrogliomas and ependyrnomas by immunohistochemistry to determine their role in diagnosis and differential diagnosis of central neurocytomas, Methods 12 cases of central neurocytomas, 20 cases of oligodendrogliomas and 20 cases of ependymomas were studied and the expression of NeuN, synaptophysin, Oligo-2 and GFAP was detected using EnVision immunohistochemistry. Results NeuN and Oligo-2 were expressed in nuclei of tumor cells while synaptophysin and GFAP in cytoplasm or membrane. 9 cases for NeuN, 10 cases for synaptophysin, 1 case for Oligo-2 and 4 cases for GFAP were positive in central neurocytomas. 2 cases for NeuN, 1 case for synaptophysin, 13 cases for Oligo-2 and 3 cases for GFAP were positive in oligodendrogliomas. 3 cases for NeuN, 2 cases for synaptophysin, 3 case for Oligo-2 and 15 cases for GFAP were positive in ependmomas.Conclusion Over-expression of NeuN and synaptophysin and lower expression of Oligo-2 and GFAP indicate that they may be valuable in diagnosis and differential diagnosis of central neurocytomas.
