摘要
目的利用RNA干扰(RNAi)技术通过热休克蛋白47(HSP47)siRNA重组质粒和脂质体的混合液对裸鼠瘢痕疙瘩模型的体内干预,分析HSP47基因在瘢痕疙瘩生成中的意义。方法构建裸鼠瘢痕疙瘩动物模型,第16天实验组(n=8)在瘢痕疙瘩内注射HSP47 siRNA重组质粒、脂质体混合液0.25 ml,对照组(n=8)裸鼠瘢痕疙瘩内注射PBS液0.25 ml。所有动物原笼饲养,7天回收标本,分别作mRNA水平、胶原蛋白水平检测及免疫组化染色。结果实验组体内干预后,HSP47的基因表达水平均较对照组明显下降(P<0.01),胶原的表达水平明显低于对照组(P<0.01)。结论HSP47基因能促进瘢痕疙瘩生成,其可能成为抑制瘢痕疙瘩的新靶点。
Objective To analyze the significance of heat shock protein 47 (HSP47) gene in keloid formation by the in vivo intervention with RNAi technology and the recombinant plasmid of HSP47 siRNA against HSP47 in nude mice keloid model. Methods RNAi mixture (0.25 ml) was injected into the keloid in experiment group and PBS (0.25 ml) was injected into the keloid in control group on the 16th day after the models were established. Following in vivo intervention, the specimens were observed and harvested on the 7th day for determination of mRNA and collagen levels and immunohistochemical staining. Results After in vivo intervention, the expression of HSP47 gene remarkably decreased in contrast to the control group ( P 〈 0.01 ), and the expression of collagen was markedly lower than the control group ( P 〈 0.01 ). Conclusion The expression of HSP47 gene promotes the formation of keloid, which provides a new target to treatment of keloid.
