阿泊拉霉素抗性基因的点突变和功能分析

摘要 链霉菌基因组中常见连续两个基因重叠的现象(如ATGA),这为基因的同框敲除或取代研究带来很大困扰。对常用的报告基因——阿泊拉霉素抗性基因的5′端进行定点突变,使之可应用于这种链霉菌重叠基因的同框取代,功能验证表明,点突变没有影响到这个报告基因的生物学活性。 The gene overlapping (ATGA) is widespread in streptomycetes, making it difficult to perform in-frame deletion or replacement for functional analysis of target genes. Here site-specific mutations were incorporated into the 5' terminal of apramycin-resistant gene, allowing it suitable for in-frame replacement of genes overlapped with upstream genes by ATGA. The result proved that apramycin-resistant gene with site mutagenesis was still functional to be used for gene replacement of overlapping genes in streptomycetes.
