摘要
通过对凝集型酵母聚凝体进行解离试验,确定该凝集型酵母具有耐机械剪切能力;用解氢键试剂尿素处理酵母聚凝体时,酵母的聚凝体发生解体。脲浓度和解离时间对菌体凝集性起着显著作用,因素作用的主次顺序为脲浓度>解离时间>玻璃珠数量,即脲浓度对菌体凝集性影响最大。正交试验得到的最优条件为脲浓度1mol/L,解离时间1h,玻璃珠数量0个。提示氢键在该酵母聚凝中可能起关键作用,其机理还有待于进一步研究。
By the dissociation test for agglutinating yeast aggregates, it indicated that the agglutinating yeast used in this study was very resistant to shearing. The aggregates of agglutinating yeast would be dissociated when it was treated by urea which could catalyze the breaking of hydrogen bonds. The urea concentration and dissociation time played an important role in the agglutination of yeast. The influence order of each factor was that the urea concentration 〉 the dissociation time 〉 the amount of beading. That is to say the urea concentration played the most important role in the agglutination. The optimal conditions were confirmed by orthogonal test as follows: urea 1 mol/L, dissociation time 1 h, without beading. The results also indicated that the hydrogen bond might play an important role in the agglutination of yeast. The mechanism need to be further investigated.
