摘要
目的:观察氧化低密度脂蛋白(ox-LDL)对血管平滑肌细胞(VSMC)血管紧张素转化酶(ACE)表达的影响及凝集素样氧化低密度脂蛋白受体-1(LOX-1)在其中的作用。方法:人脐带动脉平滑肌细胞体外原代培养,传至4~5代用于实验。ox-LDL干预,RT-PCR测定VSMC ACE mRNA和LOX-1mRNA表达。并观察予以LOX-1的抑制剂多聚肌苷酸作用后,VSMC ACE mRNA表达的变化。结果:①较高浓度ox-LDL(20mg/L,100mg/L)作用后,VSMC ACE mRNA表达升高,作用1h出现峰值。低浓度ox-LDL(10mg/L)此种作用不明显。②ox-LDL作用后,VSMCLOX-1 mRNA表达升高,且随着ox-LDL浓度的升高而表达增强。③多聚肌苷酸抑制LOX-1后,ACE mRNA表达明显下降。结论:ox-LDL明显促进VSMC ACE表达,LOX-1在此过程中起重要介导作用。
Objective: To observe the effect of oxidized low density lipoprotein (ox-LDL) and lectin-like low density lipoprotein receptor-1 (LOX-1) on angiotensin converting enzyme (ACE) mRNA expression of vascular smooth muscles cells (VSMC). Method:Smooth muscle cell from human umbilical artery was cultured in vitro and 4-5th generation was taken for use. Interfered in ox-LDL, the quantity of VSMC ACE mRNA and LOX-1 mRNA were detected by RT PCR. And the changes of VSMC ACE mRNA expression was observed after treatment of polyinosinic acid which was an inhibitor of LOX1. Result: (1)The quantity of VSMC ACE mRNA increased after co-cultured with higher concentration ox LDL(20 mg/L, 100 mg/L), and the peak value was found at about the lth hours. No obvious influence was found for low concentration ox-LDL(10 mg/L). (2)The quantity of VSMC LOX-1 mRNA increased significantly with dependence on ox-LDL concentration. (3)ACE mRNA was reduced significantly when LOX-1 was inhibited by polyinosinie acid. Conclusion:ox-LDL can induce the expression of VSMC ACE mRNA in which LOX-1 plays a vital rule.
