摘要
目的研究不同糖浓度下成纤维细胞的生长情况。方法采用体外模型,观察不同糖浓度的培养液下成纤维细胞的生长情况,对细胞增殖和Ⅰ型胶原合成情况进行研究。结果当培养液糖浓度低于40mmol/L时,成纤维细胞的增殖并未发现受到太多抑制,随着糖浓度的升高,成纤维细胞的增殖随之抑制(P〈0.01);成纤维细胞分泌Ⅰ型胶原的量与糖浓度无明显相关(P〉0.05)。随着糖浓度的升高,成纤维细胞产生肌动蛋白(α-smooth muscle actin,α-SMA)的量增加。结论应激状态下,当培养液糖浓度在一定范围内(25—40mmol/L),并不影响创面的愈合,当高于这个范围,创面的愈合将得到抑制。成纤维细胞分泌Ⅰ型胶原的量与糖浓度无直接关系;当糖浓度升高时,成纤维细胞向肌成纤维细胞转化的发生率随之升高。
Objective To find the relationship between glucose concentration and the proliferation of fibroblast. Methods The proliferation of mouse fibroblast was tested in different concentration of glucose at 12h,24h, 48h with CCK kit. The amount of the out-cell type I collagen fibers was tested by the mouse type I collagen Elisa kit,and the expression of a-smooth muscle actin (a-SMA) tested by immunochemistry. Results We found that under the medium of low concentration, the proliferation was much more than that in the high concentration groups after 12h,24h and 48h(P 〈 0.01) , namely the concentration was negatively correlated with the proliferation. We tested the out-cell type I collagen fibers using the mouse type I collagen Elisa kit, but there was no difference between each group ( P 〉 0.05 ). About the expression of mouse α-SMA, positive correlation was found between the glucose concentration and the expression of the mouse α-SMA. Conclusion At the original augmentation of the blood glucose, the proliferation of fibroblast is not different. But when it gets over a level, the proliferation will be depressed, which can lead to the delay of the healing of raw surface ; the concentration of glucose has nothing to do with the out-cell type I collagen fibers ; the concentration of glucose is a factor which enhances the variation from fibroblast to myofibroblast, and the concentration of glucose is positively correlated with the tendency of the variation.
