摘要
以牦牛胸小骨为原料,采用稀碱-酶解法提取硫酸软骨素,对碱解条件和酶解过程进行筛选.碱提取的最佳条件为:原料与1.5%的NaOH液质量比1∶1.2,38℃搅拌提取10 h;酶水解最佳条件为:胰酶体积分数8%,pH8.5~9.0,48~51℃水解5~6 h.在最佳提取条件下,得到牦牛胸小骨硫酸软骨素产品为白色粉末,纯度92.5%,收率为2.67%.
hondroitin sulfate (CS) was extracted from yak breastbone by using sparse alkali-enzymatic decompounding method. Some technique conditions of alkali extracting and enzymatic decompounding were screened by orthogonal experiment. The results showed that the optimum condition for alkali extracting was 1.5% NaOH as extraction solvent, the ratio of material to solvent 1 : 1.2, controlling temperature at 38 ℃ and stirring for 10 hours, The optimum condition for enzymatic decompounding was trypsinogen 8 %,temperature 48-51 ℃, pH 8.5 - 9.0 and reaction time 5-6 hours. Under above extracting conditions,CS was obtained as white powder,with 92.5 % pure degree and 2.67% of extracting rate.
